● 產(chǎn)品簡介:

本產(chǎn)品是由6條雙鏈DNA條帶組成,適用于瓊脂糖凝膠電泳中DNA條帶的分析。產(chǎn)品中已含有1×loading buffer,可根據(jù)實(shí)驗(yàn)需要,直接取2-5 μl電泳,使用方便,電泳圖像清晰。6條帶的大小分別為100,250,500,750,1000,2000 bp。其中750 bp條帶濃度約為100 ng/5 μl,顯示加亮帶,其余條帶濃度約為50 ng/ 5μl。

● 貯存、效期及運(yùn)輸:

-20℃貯存;有效期3年;濕冰運(yùn)輸。

● 使用方法:

1. 取2-5 μl本產(chǎn)品加入到瓊脂糖凝膠的加樣孔中(每1 mm×1 mm加樣孔加1 μl,如果加樣孔

寬于5 mm,可以適當(dāng)增加上樣量)就行電泳。

2. 建議電泳條件:凝膠濃度為1.0-2.0%,凝膠長度5-7 cm,電泳電壓4-10 v/cm,電泳時(shí)間15-20分鐘。

3. 通過核酸染料染色后紫外燈或藍(lán)光儀觀察條帶。

● 注意事項(xiàng):

1. 瓊脂糖的質(zhì)量對(duì)DNA的電泳有很大影響,電泳時(shí)請(qǐng)盡量使用質(zhì)量優(yōu)等的瓊脂糖。

2. 由于PAGE電泳的特殊性,該產(chǎn)品不建議用于PAGE電泳。

3. 請(qǐng)使用新鮮配制的電泳緩沖液和新鮮配制的凝膠進(jìn)行電泳,以保證Marker良好的分離效果。

4. 該Marker適用于DNA片段大小的確定和對(duì)DNA含量的粗略定量,不適用于DNA片段含量的精確定量。

5.推薦使用RealSafe系列染料(貨號(hào):GR001,GR002,GR003,GR004)染色。如果使用Goldview(貨號(hào):GV001)或EB(貨號(hào):EB001)等染料進(jìn)行染色,由于此類染料帶更多的正電荷(染料移動(dòng)方向與核酸泳動(dòng)方向相反),隨著電泳時(shí)間延長,染料向大片段聚集,凝膠會(huì)出現(xiàn)陰陽膠現(xiàn)象(染料含量高的凝膠紫外下較亮,含量低的凝膠紫外下發(fā)暗),導(dǎo)致小分子

量DNA顯色很弱或不可見。陰陽膠可以在電泳結(jié)束后浸泡染色,將膠浸沒在含EB或Goldview的電泳緩沖液中搖動(dòng)染色20-25 min,小分子量DNA會(huì)顯色清楚。更長時(shí)間浸泡會(huì)因DNA分子擴(kuò)散而使條帶彌散,小分子量DNA更易彌散。

6.紫外照射下EB易分解,使DNA條帶熒光變淺。DNA受紫外光照射形成嘧啶二聚體,不利于待回收片段的下游工作,因此盡可能減少紫外照射時(shí)間。

7.蔗糖會(huì)結(jié)合DNA,降低其電泳遷移率,建議DNA樣品上樣使用含甘油的上樣緩沖液,如6×DNA上樣緩沖液(雙染料)(貨號(hào):DL020)。

8.大部分核酸工具酶會(huì)結(jié)合DNA,降低其電泳遷移率,大分子量DNA尤為明顯,建議此類DNA樣品上樣使用含SDS的上樣緩沖液,如6×DNA上樣緩沖液(含SDS)(貨號(hào):DL090)。
